Article Synopsis
  • Metabolic flexibility in skeletal muscle is crucial for healthy glucose and lipid metabolism, and its dysfunction can lead to metabolic diseases.
  • Exercise improves metabolic flexibility and helps identify mechanisms that support metabolic health.
  • The study reveals that pantothenate kinase 4 (PanK4) is vital for muscle metabolism, as its deletion disrupts fatty acid oxidation and elevates harmful acetyl-CoA levels, which lead to glucose intolerance, while increasing PanK4 enhances glucose uptake and lowers acetyl-CoA.

  • Distal sensorimotor polyneuropathy (DSPN) is a prevalent neurological condition affecting older adults and those with obesity or diabetes, leading to significant health issues.
  • The Interpretable Multimodal Machine Learning (IMML) framework was used to predict the prevalence and incidence of DSPN by analyzing a diverse set of data from over 1,000 participants, including clinical, genomic, and metabolomic information.
  • Results showed that while clinical data alone could differentiate DSPN cases, combining it with additional molecular data improved prediction accuracy and identified potential biomarkers related to inflammation and fatty acid metabolism, offering new insights for treatment and prevention strategies.

Introduction: In response to the COVID-19 pandemic, we rapidly implemented a plasma coordination center, within two months, to support transfusion for two outpatient randomized controlled trials. The center design was based on an investigational drug services model and a Food and Drug Administration-compliant database to manage blood product inventory and trial safety.

Methods: A core investigational team adapted a cloud-based platform to randomize patient assignments and track inventory distribution of control plasma and high-titer COVID-19 convalescent plasma of different blood groups from 29 donor collection centers directly to blood banks serving 26 transfusion sites.

The use of normothermic machine perfusion (NMP) over static cold storage in liver transplantation has been shown to reduce posttransplant risks of early allograft dysfunction, primary nonfunction, and ischemic cholangiopathy, and its increasing use has played a role in the expanded utilization of marginal livers. While studies have demonstrated improved clinical outcomes using NMP over static cold storage preservation, real-time intraoperative data reflecting the quality and viability of NMP livers is limited. This retrospective, single-center study compared NMP versus static cold storage livers in first-time recipients of liver transplants through the evaluation of synthetic coagulation function as measured by thromboelastography and conventional coagulation testing.

Metabolomics and lipidomics are pivotal in understanding phenotypic variations beyond genomics. However, quantification and comparability of mass spectrometry (MS)-derived data are challenging. Standardised assays can enhance data comparability, enabling applications in multi-center epidemiological and clinical studies.

Background: Sex differences exist in type 2 diabetes (T2D), and androgens have been implicated in the etiology of T2D in a sex-specific manner. We therefore aimed to investigate whether androgens play a role in explaining sex differences in glucose homeostasis and incidence of T2D.

Methods: We used observational data from the German population-based KORA F4 study (n = 1975, mean age: 54 years, 41% women) and its follow-up examination KORA FF4 (median follow-up 6.

Purpose: Autologous serum tears (ASTs) are produced from serum obtained by blood draw and used to treat severe dry eye diseases. Multiple cases of failed AST production occurred due to serum becoming a solidified, gel-like substance after centrifugation. The aim of this study was to identify risk factors associated with inability to compound AST.

Article Synopsis
  • Type 2 diabetes (T2D) and obesity are linked to low levels of natriuretic peptide (NP) and reduced NP guanylyl cyclase receptor-A (GCA) in muscles and fat tissue.
  • Research in mice shows that lack of ANP/GCA leads to metabolic issues and prediabetes, causing insulin resistance and poor endurance.
  • ANP/GCA is crucial for maintaining mitochondrial function and oxidative capacity in skeletal muscle, suggesting it plays a key role in the development of prediabetes.

Article Synopsis
  • Biliary atresia (BA) is a rare liver disease in newborns that often goes unnoticed at first.
  • Researchers in Germany looked at blood samples from newborns to see if they had different amino acids if they later got diagnosed with BA.
  • They discovered that some babies with BA had certain amino acids at higher levels shortly after birth, suggesting the disease may start before they show symptoms, which could help in early detection.

Article Synopsis
  • Researchers are studying type 2 diabetes, which happens when there is too much sugar in the blood, to see how certain substances in the body, called metabolites, are connected to it.
  • They looked at 3,000 blood samples and analyzed 911 metabolites to find out how these substances relate to blood sugar levels.
  • They discovered several metabolites that are different in people with normal blood sugar, those with prediabetes, and those with type 2 diabetes, mainly focusing on specific amino acids and fats.

Introduction/objectives: Changes in the stool metabolome have been poorly studied in the metabolic syndrome (MetS). Moreover, few studies have explored the relationship of stool metabolites with circulating metabolites. Here, we investigated the associations between stool and blood metabolites, the MetS and systemic inflammation.

Context: The independent role of glomerular filtration rate (GFR) decline in shaping the risk of mortality in people with type 2 diabetes has only been partially addressed.

Objective: The objective of the study was twofold: i) to investigate the association between all-cause mortality and eGFR changes over time; ii) to understand whether renal dysfunction mediates the effect of tryptophan metabolism on death risk.

Design: Prospective study with an average follow-up of 14.

This study introduces an orbital monitoring system designed to quantify non-technical losses (NTLs) within electricity distribution networks. Leveraging Sentinel-2 satellite imagery alongside advanced techniques in computer vision and machine learning, this system focuses on accurately segmenting urban areas, facilitating the removal of clouds, and utilizing OpenStreetMap masks for pre-annotation. Through testing on two datasets, the method attained a Jaccard index (IoU) of 0.

Choroid plexus carcinomas (CPC) are early childhood cancers characterized by loss of function and poor survival. We are analyzing data on status, survival, and second cancers from the largest cohort of CPC receiving chemotherapy followed by consolidation with marrow-ablative chemotherapy (HDCx). Additionally, we discuss the rationale for targeted therapies for CPC patients.

Research has indicated that sex hormone-binding globulin (SHBG) is associated with glucose homeostasis and may play a role in the etiology of type 2 diabetes (T2D). While it is unclear whether SHBG may mediate sex differences in glucose control and subsequently, incidence of T2D. We used observational data from the German population-based KORA F4 study (n = 1937, mean age: 54 years, 41% women) and its follow-up examination KORA FF4 (median follow-up 6.

Background: Metformin and sodium-glucose-cotransporter-2 inhibitors (SGLT2i) are cornerstone therapies for managing hyperglycemia in diabetes. However, their detailed impacts on metabolic processes, particularly within the citric acid (TCA) cycle and its anaplerotic pathways, remain unclear. This study investigates the tissue-specific metabolic effects of metformin, both as a monotherapy and in combination with SGLT2i, on the TCA cycle and associated anaplerotic reactions in both mice and humans.

Combining a Sodium-Glucose-Cotransporter-2-inhibitor (SGLT2i) with metformin is recommended for managing hyperglycemia in patients with type 2 diabetes (T2D) who have cardio-renal complications. Our study aimed to investigate the metabolic effects of SGLT2i and metformin, both individually and synergistically. We treated leptin receptor-deficient (db/db) mice with these drugs for two weeks and conducted metabolite profiling, identifying 861 metabolites across kidney, liver, muscle, fat, and plasma.
