Cell therapy with human IL-10-producing ILC2s limits xenogeneic graft-versus-host disease by inhibiting pathogenic T cell responses.

Interleukin-10 (IL-10)-producing group 2 innate lymphoid cells (ILC2) regulate inflammatory immune responses, yet their therapeutic potential remains largely unexplored. Here, we demonstrate that cell therapy with human ILC2 inhibits pathogenic T cell responses in humanized mouse models of graft-versus-host disease (GVHD), resulting in reduced GVHD severity and improved overall survival without limiting the graft-versus-leukemia effect. ILC2 conferred superior protection from GVHD than IL-10 ILC2s, and blocking IL-10 and IL-4 abrogated ILC2 protective effects, indicating that these cytokines are important for the protective effects of ILC2.

Experience-based theories of language processing suggest that listeners use the properties of their previous linguistic input to constrain comprehension in real time (e.g. MacDonald & Christiansen, 2002; Smith & Levy, 2013; Stanovich & West, 1989; Mishra, Pandey, Singh, & Huettig, 2012).

Single-cell profiling of healthy human kidney reveals features of sex-based transcriptional programs and tissue-specific immunity.

Knowledge of the transcriptional programs underpinning the functions of human kidney cell populations at homeostasis is limited. We present a single-cell perspective of healthy human kidney from 19 living donors, with equal contribution from males and females, profiling the transcriptome of 27677 cells to map human kidney at high resolution. Sex-based differences in gene expression within proximal tubular cells were observed, specifically, increased anti-oxidant metallothionein genes in females and aerobic metabolism-related genes in males.

Immunoglobulin A nephropathy is characterized by anticommensal humoral immune responses.

IgA nephropathy (IgAN) is a leading cause of kidney failure, yet little is known about the immunopathogenesis of this disease. IgAN is characterized by deposition of IgA in the kidney glomeruli, but the source and stimulus for IgA production are not known. Clinical and experimental data suggest a role for aberrant immune responses to mucosal microbiota in IgAN, and in some countries with high disease prevalence, tonsillectomy is regarded as standard-of-care therapy.

Individual differences in syntactic processing: Is there evidence for reader-text interactions?

There remains little consensus about whether there exist meaningful individual differences in syntactic processing and, if so, what explains them. We argue that this partially reflects the fact that few psycholinguistic studies of individual differences include multiple constructs, multiple measures per construct, or tests for reliable measures. Here, we replicated three major syntactic phenomena in the psycholinguistic literature: use of verb distributional statistics, difficulty of object-versus subject-extracted relative clauses, and resolution of relative clause attachment ambiguities.

Visualization of 3D White Adipose Tissue Structure Using Whole-mount Staining.

Adipose tissue is an important metabolic organ with high plasticity and is responsive to environmental stimuli and nutrient status. As such, various techniques have been developed to study the morphology and biology of adipose tissue. However, conventional visualization methods are limited to studying the tissue in 2D sections, failing to capture the 3D architecture of the whole organ.

Language comprehension requires successfully navigating linguistic variability. One hypothesis for how listeners manage variability is that they rapidly update their expectations of likely linguistic events in new contexts. Peri-operative Levosimendan in Patients Undergoing Cardiac Surgery: An Overview of the Evidence.

Levosimendan, a calcium sensitiser, has recently emerged as a valuable agent in the peri-operative management of cardiac surgery patients. Levosimendan is a calcium-sensitising ionodilator. By binding to cardiac troponin C and reducing its calcium-binding co-efficient, it enhances myofilament responsiveness to calcium and thus enhances myocardial contractility without increasing oxygen demand.

An assessment of WISC-IIIUK on children with HIV infection.

The Wechsler Intelligence Scale for Children - Third Edition UK test was administered to groups of children between the ages of 6 and 12 years with vertically transmitted HIV infection (n = 70) and a control group who were not infected by the virus (n = 70). The study was conducted in India. The two groups were matched for general verbal abilities, age and gender.

Verbal working memory predicts co-speech gesture: evidence from individual differences.

Gesture facilitates language production, but there is debate surrounding its exact role. It has been argued that gestures lighten the load on verbal working memory (VWM; Goldin-Meadow, Nusbaum, Kelly, & Wagner, 2001), but gestures have also been argued to aid in lexical retrieval (Krauss, 1998). In the current study, 50 speakers completed an individual differences battery that included measures of VWM and lexical retrieval.

Spatially distributed scintillator arrays for diagnosing runaway electron transport and energy behavior in tokamaks.

We present details of a new bismuth germanate [Bi(4)Ge(3)O(12) (BGO)] scintillator array used to diagnose the transport and energy behavior of runaway electrons (REs) in DIII-D. BGO exhibits important properties for these compact detectors including high light yield which sufficiently excites photodiode detectors (8500 photons/MeV), high density and atomic numbers of constituent materials which maximizes sensitivity, and relative neutron blindness which minimizes complications in data interpretation. The detectors observe primarily hard x-ray radiation emitted in a forward beamed pattern by RE when they strike first wall materials or bulk ions and neutrals in the plasma, although we also address photoneutron signals.

Sm(II) reduction chemistry of heteroalkynes: stable adducts, reductive coupling, reductive C-C/C-N bond cleavage and trapping of the tert-butyl fragment with bulky nitriles, phosphaalkynes and isonitriles.

Reactions of a dimetallated N,N'-dimethyl substituted porphyrinogen Sm(II) complex with a series of t-butyl substituted heteroalkynes affords a diverse range of reactivity. The phosphaalkyne t-BuC[triple bond]P gives a dinuclear Sm(III) P-P reductively coupled complex of (t-BuC=PP=C-t-Bu)(2-) featuring a new mu-eta(2)(1,2-C,P) binding mode. In contrast, the nitrile aza analogue t-BuC[triple bond]N forms Sm(II) adducts that undergo reductive C-C bond cleavage at elevated temperatures to afford a trimeric Sm(III) cyanide (mu-C[triple bond]N(-)) complex.

Goal-directed decision making in prefrontal cortex: A computational framework.

Research in animal learning and behavioral neuroscience has distinguished between two forms of action control: a habit-based form, which relies on stored actio n values, and a goal-dir ected form, which forecasts and compares action outcomes based on a model of the environment. While habit-based control has been the subject of extensive computational research, the computational principles underlying goal-directed control in animals have so far received less attention. In the present paper, we advance a computational framework for goal-directed control in animals and humans.

Blastocyst development rate impacts outcome in cryopreserved blastocyst transfer cycles.

Objective: To assess cycle outcome among day 5 and day 6 cryopreserved frozen-thawed blastocyst embryo transfers (FBET).

Design: Retrospective cohort study.

Setting: Military-based assisted reproduction technology (ART) center.

Quenching of the nonlocal electron heat transport by large external magnetic fields in a laser-produced plasma measured with imaging thomson scattering.

We present a direct measurement of the quenching of nonlocal heat transport in a laser-produced plasma by applying large external magnetic fields (>10 T). The temporally resolved Thomson-scattering measurements of the electron temperature profile show that the heat front propagation transverse to a high-power laser beam is slowed resulting in extremely strong local heating. We find agreement with hydrodynamic modeling when including a magnetic field model that self-consistently evolves the fields in the plasma.

Ocular syphilis in HIV-positive patients receiving highly active antiretroviral therapy.

Background: From October 2001 to October 2002, we have observed a surprisingly high incidence of ocular syphilis in human immunodeficiency virus-positive (HIV+) patients receiving highly active antiretroviral therapy at our clinic.

Methods: We conducted a retrospective chart and patient database review.

Results: From 1997 to 2002, 455 patients in our clinic were screened for syphilis; 320 were screened from 2001 to 2002; 7.

The limited importance of pronuclear scoring of human zygotes.

Background: Several studies have shown a correlation between the pronuclear morphology score (PNMS) and subsequent embryo development and implantation. Embryos with poor pronuclear score, elsewhere referred to as Z3 and Z4, are often not transferred or cryopreserved because it is believed that they have poor pregnancy potential. The objective of this study is to report our data on the use of the pronuclear score and its effect on pregnancy outcome.

Reporting of deaths during pre-approval clinical trials for advanced HIV-infected populations.

The Division of Antiviral Drug Products of the US FDA has regulatory authority over the investigational new drugs under development by various sponsors to treat HIV-infected populations. The FDA and the sponsors of investigational new drugs use the Code of Federal Regulations to guide the entire drug development process, in order to ensure that safe and efficacious drugs are brought to market. To achieve this goal, diligent monitoring for safety during the pre-approval phase of new drug development is particularly crucial.
