In mass spectrometry-based proteomics, loss-minimized peptide purification techniques play a key role in improving sensitivity and coverage. We have developed a desalting tip column packed with thermoplastic polymer-coated chromatographic particles, named ChocoTip, to achieve high recoveries in peptide purification by pipet-tip-based LC with centrifugation (tipLC). ChocoTip identified more than twice as many peptides from 20 ng of tryptic peptides from Hela cell lysate compared to a typical StageTip packed with chromatographic particles entangled in a Teflon mesh in tipLC.

To understand the biological and pathological functions of protein glycosylation, the glycan heterogeneities for each glycosite in a single glycoprotein need to be elucidated depending on the type and status of cells. For this aim, a reliable strategy is needed to compare site-specific glycoforms of multiple glycoprotein samples in a comprehensive manner. To analyze this "inter-heterogeneity" of samples, we previously introduced an MS1-based glycopeptide detection method, "Glyco-RIDGE.

Article Synopsis
  • A 71-year-old man with metastatic hormone-sensitive prostate cancer was treated with a combination of medications and palliative radiation therapy targeting specific bones.
  • Following the treatment, he developed severe neutropenia on day 35, which halted his treatment and led to a recovery period of 23 days using a granulocyte colony-stimulating factor.
  • The case highlights the critical need to prioritize treatment by focusing on specific lesions and to avoid irradiating areas rich in red bone marrow to prevent severe hematological toxicity.
