Objective: Plasma metabolite profiling has uncovered several nonglycemic markers of incident type 2 diabetes (T2D). We investigated whether such biomarkers provide information about specific aspects of T2D etiology, such as impaired fasting glucose and impaired glucose tolerance, and whether their association with T2D risk varies by race.

Research Design And Methods: Untargeted plasma metabolite profiling was performed of participants in the FINRISK 2002 cohort (n = 7,564).

Article Synopsis
  • - The study investigates the link between insulin resistance and various metabolic diseases, emphasizing the need to consider insulin resistance in epidemiological studies.
  • - By analyzing genetic data from over 382,000 individuals, researchers identified 251 genetic loci related to insulin resistance and prioritized key genes through further analysis.
  • - Notable findings include genes like PLA2G12A and PLA2G6, which enhance insulin sensitivity, and TNFAIP8, which affects insulin resistance differently in females by increasing fat accumulation; these could be potential biomarkers or drug targets.

Article Synopsis
  • Loss-of-function mutations in the HNF1A gene are linked to rare diabetes forms and affect liver biology, but mechanisms remain unclear.
  • A study analyzed nearly 12,000 protein variants in liver cells and compared them to genetic data from over half a million individuals to explore their effects.
  • Remarkably, about 1 in 5 rare variants actually increase HNF1A's function, protecting carriers from type 2 diabetes, but also promoting liver-related conditions by raising harmful fat levels in the blood.

Purpose: To investigate the real-world safety of paclitaxel (PTX)-coated devices for treating lower extremity peripheral artery disease using a commercial claims database.

Materials And Methods: Data from FAIR Health, the largest commercial claims data warehouse in the United States, were used for this study. The study consisted of patients who underwent femoropopliteal revascularization procedures between January 1, 2015, and December 31, 2019, with PTX and non-PTX devices.

Adipose tissue plays a crucial role in maintaining metabolic homeostasis by storing lipids and glucose from circulation as intracellular fat. As peripheral tissues like adipose tissue become insulin resistant, decompensation of blood glucose levels occurs causing type 2 diabetes (T2D). Currently, modulating the glycocalyx, a layer of cell-surface glycans, is an underexplored pharmacological treatment strategy to improve glucose homeostasis in T2D patients.
