In this study, we introduce a new method for oligonucleotide fragment assembly. Unlike polymerase chain assembly and ligase chain assembly that rely on short, highly purified oligonucleotides, our method, named , uses a one-tube, splint-driven assembly reaction. Splynthesis connects standard-desalted "contig" oligos (∼150 nt in length) via shorter "splint" oligos harboring 5' and 3' blocking modifications to prevent off-target ligation and amplification events.

The introduction of a large predatory snow crab, , into the Kara Sea is a unique situation where the impact of an invasive species affecting an otherwise undisturbed ecosystem can be observed unhindered by other ecosystem stressors. Trophic interactions are one of the principal relationships between animals and can help assess an ecosystem's stability. The trophic positions and sources of organic material for the most common benthic species of Blagopoluchiya Bay sampled at different stages of the invasion were calculated using stable isotope analysis.

In the present study, we demonstrated that the introduction of a 1,4-diethyl-1,2,3,4-tetrahydroquinoxalin moiety into the arylidene part of GFP chromophore-derived compounds results in the formation of environment-sensitive fluorogens. The rationally designed and synthesized compounds exhibit remarkable solvent- and pH-dependence in fluorescence intensity. The solvent-dependent variation in fluorescence quantum yield makes it possible to use some of the proposed compounds as polarity sensors suitable for selective endoplasmic reticulum fluorescent labeling in living cells.
