Background: Somatostatin analogues have been used successfully for the treatment of acromegaly but no randomized studies have compared the effects of lanreotide Autogel (LAN) and octreotide acetate long-acting repeatable (OCT).
Objective: To compare the effect of LAN and OCT for the treatment of acromegaly in a randomized study design.
Material And Methods: Twelve acromegalic patients were included and 10 patients completed treatment with LAN or OCT for 6 months and were then switched to the opposite treatment modality for 6 months without a washout period in a randomized crossover design.

Of 605 hyperprolactinaemic sera (prolactin > 1000 mU l-1 determined with PRL DELFIA, Wallac) in the routine diagnostic (PRL was measured in 10,737 sera) 26% had an increased amount of PRL with molecular weight (MW) = 150-170 kD (150-170 kD PRL or bigbig PRL). In a series of serum samples from 660 healthy subjects, only one female with hyperprolactinaemia due to increased 150-170 kD PRL was found. The 150-170 kD PRL constituted less than 1% of the total PRL found in 11 patients with prolactinoma (macroprolactinoma, PRL 8800-500,000 mU l-1).
View Article and Find Full Text PDFThe long-term efficacy and tolerability of CV 205-502, a non-ergot dopamine agonist with D-2 receptor affinity, were studied for up to 36 months in 16 patients with macroprolactinomas. Prolactin values were reduced in all cases, becoming either normalized or suppressed in 12. The pituitary tumor size was reduced in the 13 patients with an obvious tumor and visual function normalized in all six patients with initial defects.
View Article and Find Full Text PDFTwenty-six acromegalic patients were randomized to treatment with either SMS 201-995 or bromocriptine in increasing doses and were investigated before treatment, after 2, 4, and 8 weeks of treatment, and 2 weeks after discontinuation of treatment. There were two dropouts from the bromocriptine group and one from the SMS 201-995 group. Amelioration of clinical signs and symptoms was seen in both groups during treatment.
