The leukemia is a neoplastic clonal disease of hematopoietic system with primary affection of bone marrow. The modern technologies of automated blood analysis permit to implement a quick primary screening of pathological samples of blood suspicious for presence of blast cells. The application of various analysis techniques (optical, cytochemical, immune phenotypical) with the purpose of detecting blast cells demonstrates their different distribution at the graphics.

The article discusses the parameters of evaluation of thrombocytes activity detected by hematologic analyzers. The techniques of parameters detection are presented using such devices as XE-5000, XE-2100 (Sysmex), LH750, 780, UniCel DxHTM 800 (Beckman Coulter), Advia 120, 2120 (Siemens). The dependence of normal values of these indicators from age and gender are discussed.

The paper reviews literature updates on the examination of platelets by the flow cytofluorometric technique; it analyzes in detail the effect of various anticoagulants used to stabilize blood, the time and conditions of sample storage at the preanalytical stage of studies on the results of an analysis. Different platelet subpopulations detected by the flow cytofluorometric technique are described and their clinical value is shown. The clinical importance of reticular platelet tests is separately considered.

The paper reviews literature updates on the organization and specific features of megakaryocytopoiesis, the mechanisms of its regulation, possible ways for platelets to occur from megakaryocytes. It characterizes thrombocytopoiesis, by describing the detailed structural organization of platelets and gives data on the relationship of the morphofunctional features of platelets to the ploidy of megakaryocytes that give rise to the latter. The structure and clinical value of a number of platelet activation markers, such as CD41/CD61, CD42, CD62p, platelet-leukocyte aggregates, microvesicles, are described.
