Article Synopsis
  • A(H5Nx) avian influenza viruses are widespread and evolving, with a notable emergence of highly pathogenic A(H5N1) clade 2.3.4.4b reassortant viruses since 2020.
  • Some of these viruses, particularly those isolated from mammals, carry a mutation (E627K) in the PB2 protein that enables adaptation to mammalian cells.
  • Recent findings from Russia reveal a highly pathogenic A(H5N1) virus with the E627K mutation that shows increased virulence in mice and limited airborne transmission in ferrets, underscoring the need for ongoing surveillance to mitigate pandemic risks.

Article Synopsis
  • - The study focuses on correcting mutations in the CASR gene, which is crucial for regulating calcium levels in the body, specifically targeting mutations linked to a severe condition called neonatal primary hyperparathyroidism.
  • - Researchers generated a CRISPR/Cas9 edited human induced pluripotent stem cell (hiPSC) line that successfully restored one of the mutations while preserving another, showing normal characteristics such as cell structure and growth potential.
  • - The newly created hiPSC line will be used as a valuable resource for understanding calcium regulation disorders and exploring personalized medicine approaches to treat these conditions in the future.

Anti-human immunodeficiency virus (HIV) broadly neutralizing antibodies (bNAbs) offer a promising approach for the treatment of HIV-1. The current paradigm for antibody therapy involves passive antibody transfer, requiring regular delivery of bNAbs in treating chronic diseases such as HIV-1. An alternative strategy is to use AAV-mediated gene transfer to enable in vivo production of desirable anti-HIV-1 antibodies.

The lack of oxygen (O2) causes changes in the cell functioning. Modeling hypoxic conditions in vitro is challenging given that different cell types exhibit different sensitivities to tissue O2 levels. We present an effective in vivo platform for assessing various tissue and organ parameters in Danio rerio larvae under acute hypoxic conditions.

We conducted a study on the impact of intraperitoneal injections of melatonin and its three bioisosteres (compounds 1-3) on the development of oxygen-induced retinopathy in newborn rats during a 21-day experiment. It was demonstrated that melatonin and its analogues 1-3 effectively reduce the total protein concentration in the vitreous body of rat pups, decrease concentration of VEGF-A, and lower the level of oxidative stress (as indicated by normalization of antioxidant activity in the vitreous body). Melatonin and its analogues 1-3 equally normalize the level of VEGF-A.
