Morning glory syndrome (MGS) is a rare congenital optic disc malformation characterized by a distinctive funnel-shaped optic disc resembling a blooming morning glory flower, typically diagnosed in childhood or adolescence and primarily presenting unilaterally, although bilateral cases have been reported. The anomaly features an enlarged optic disc, radiating blood vessels, and surrounding excavation, leading to visual impairment that can range from mild to profound. The etiology of MGS is not fully understood but has been associated with genetic factors, particularly mutations in the PAX6 and PAX2 genes, which play critical roles in eye development.

Single-molecule localization microscopy (SMLM) is a powerful super-resolution imaging technique that offers resolution far beyond the optical diffraction limit. The commonly used high numerical-aperture (NA) objective lenses in SMLM can only provide a nearly ideal point-spread function (PSF) at the center of the field-of-view (FOV), whereas the off-axis PSF is often distorted due to optical aberrations. Since precision and accuracy of three-dimensional (3D) spatial localization of single molecules heavily depend on the system's PSF, the FOV of 3D SMLM is often restricted to about 50 µm × 50 µm limiting its applications in visualizing intra-/intercellular interactions and high-throughput single-molecule analysis.

Yeast extracellular proteases play a key role in developing the taste of dry-cured ham, whereas the mechanism of yeast proteases on taste formation of dry-cured ham is not fully studied. The proteases characteristics of yeast isolated form Jinhua ham, hydrolysis capacities for myofibrillar proteins (MP), free amino acid contents, metabolite compositions, taste parameters and the relationship between metabolites and taste parameters were investigated to reveal the mechanism of Rhodotorula mucilaginosa AUMC 9298 (RM) and Candida parapsilosis d70a (CP) proteases on MP hydrolysis and taste development of dry-cured ham. Kirsten rat sarcoma viral oncogene homolog (KRAS) oncogenic mutations are genetic drivers in various cancers, including non-small cell lung cancer (NSCLC). However, the regulatory mechanisms underlying the progression of NSCLC driven by oncogenic KRAS mutants are incompletely understood. Here, we show that ubiquitin specific peptidase 25 (USP25) impedes ring finger protein 31 (RNF31)-mediated linear ubiquitination of KRAS oncogenic mutants (KRAS) independently of its deubiquitinase activity, which facilitates the plasma membrane (PM) localization and the downstream oncogenic signaling of KRAS.

Over 90% of gastrointestinal stromal tumors (GISTs) harbor mutations in KIT or PDGFRA that can predict response to tyrosine kinase inhibitor (TKI) therapies, as recommended by NCCN (National Comprehensive Cancer Network) guidelines.
