The complex tumor microenvironment (TME) presents significant challenges to the development of effective therapies against solid tumors, highlighting the need for advanced in vitro models that better recapitulate TME biology. To address this, we developed a vascularized human liver tumor model using a microfluidic platform, designed to test both drug and cell-based therapies. This model mimics critical tumorigenic features such as hypoxia, extracellular matrix (ECM), and perfusable vascular networks.

A minimally invasive analysis of plasma cell-free DNA (cfDNA) offers a genomic profiling of early-stage breast cancer (EBC), potentially identifying mutational signatures linked to metastasis and therapy resistance. In this study, paired plasma and tissue samples from 40 hormone receptor-positive (HR+) EBC patients were sequenced using a custom-designed comprehensive gene panel, OncoIndx. The genomic landscape of circulating tumor DNA (ctDNA) showed a broader mutation spectrum compared to tumor tissue DNA (tDNA), and provided reliable assessments of microsatellite instability (MSI), tumor mutation burden (TMB), homologous recombination deficiency (HRD), and loss of heterogeneity (LOH), all indicating high genomic instability.

Background: A hybrid nerve sheath tumor is a biphasic, benign neoplasm of peripheral nerve sheaths, consisting of combinations of neurofibroma, schwannoma, or perineurioma. These tumors were recognized only recently, in 2013; they commonly occur sporadically but rarely with syndromic associations, such as neurofibromatosis syndrome, Carney complex, and schwannomatosis. With an occurrence of 1 in every 33,000 individuals, neurofibromatosis type 2 is a rare autosomal dominant condition characterized by bilateral vestibular schwannomas.

Background: Glioblastoma (GBM) carries a poor prognosis, and new therapeutic strategies are necessary to improve outcomes for patients with this disease. Alkylating chemotherapies including temozolomide (TMZ) and lomustine (CCNU) are critical for treating GBM, but resistance mechanisms, including hypomethylation of O6-methylguanine-DNA methyltransferase (MGMT) promoter, undermine treatment. CRISPRoff is a programmable epigenetic memory editor that can induce stable and heritable gene silencing after transient delivery, and we hypothesize that CRISPRoff could potentiate the activity of TMZ and CCNU through long term suppression of target genes.

IgA vasculitis (IgAV), also called Henoch-Schönlein purpura (HSP), is an immune complex vasculitis characterised by IgA1-dominant immune deposits affecting small vessels (predominantly capillaries, venules or arterioles). It often involves the skin and gastrointestinal tract, frequently causes arthritis and rarely causes vasculitis in the kidney indistinguishable from IgA nephropathy. It usually presents as a purpuric non-blanching rash mostly on the lower limbs, although it may extend to the upper limbs and the trunk, and more rarely to the face.

Targeted protein degradation (TPD) has garnered appreciable interest in drug discovery due to its unique mechanism of action - degradation of a target in an event-driven manner, instead of traditional occupancy-driven inhibitor-based therapies. This is achieved by employing mono- or hetero-bifunctional small molecules known as degraders to induce the proximity of two proteins: a target protein and an E3 ubiquitin ligase, ultimately resulting in clearance of the target protein by the cell's inherent degradation machinery. A critical step in this pathway is ternary complex formation (TCF) between the ligase, degrader molecule, and the target protein.

Objective: Tumor mutational burden (TMB) has been proposed as a prognostic biomarker in patients with metastatic cancer, as well as in patients who receive immune checkpoint inhibitor (ICI) therapy. However, the role of TMB as a biomarker for progression after resection of brain metastases, as well as perioperative ICI treatment response, is less defined. This study examined the impact of TMB on local CNS progression events in patients who underwent resection of a brain metastasis, as well as in those who received postoperative ICI treatment.
