Introduction: The COVID-19 (COronaVIrus Disease-2019) pandemic highlighted the importance of assessing the rationales behind vaccine hesitancy for the containment of pandemics. In this nationwide study, representative of the Luxembourgish population, we identified hesitant groups from adolescence to late adulthood and explored motivations both for and against vaccination.

Methods: We combined data collected via online surveys for the CON-VINCE (COvid-19 National survey for assessing VIral spread by Non-affected CarriErs) study, 1865 respondents aged 18-84, and for the YAC (Young people And Covid-19) study, 3740 respondents aged 12-29.

Nucleosomes are fundamental units of chromatin in which a length of genomic DNA is wrapped around a histone octamer spool in a left-handed superhelix. Large-scale nucleosome maps show a wide distribution of DNA wrapping lengths, which in some cases are tens of base pairs (bp) shorter than the 147 bp canonical wrapping length observed in nucleosome crystal structures. Here, we develop a thermodynamic model that assumes a constant free energy cost of unwrapping a nucleosomal bp.

The pericardium plays an important role in mechanical interactions between the right (RV) and left (LV) ventricles, referred to as ventricular interdependence. However, the exact mechanisms of its supportive role remain unknown. The present study aimed to evaluate specifically ventricular interdependence in a model of isolated biventricular working heart of large mammal, which is in absence of neurohormonal influence or series interactions, and to evaluate the impacts of intact pericardium on this phenomenon.

Article Synopsis
  • Serum HBV-RNA (seRNA) is being explored as a marker for measuring the activity of HBV replication in liver cells, and the study investigates its behavior under a specific treatment regimen.
  • The trial involved 40 individuals with chronic HBV who received a combination of tenofovir, pegylated interferon, and a nucleic-acid polymer (NAP) over 48 weeks, revealing unique seRNA patterns in response to the treatments.
  • Results showed that having a lower seRNA half-life was linked to achieving a partial or functional cure, suggesting that faster inactivation of cccDNA – the genetic material driving the virus – might lead to better treatment outcomes.

Hormone-receptor-positive (HR) luminal cells largely mediate the response to estrogen and progesterone during mammary gland morphogenesis. However, there remains a lack of consensus on the precise nature of the precursor cells that maintain this essential HR lineage. Here we refine the identification of HR progenitors and demonstrate their unique regenerative capacity compared to mature HR cells.

Scope: The emergence of post-COVID-19 condition (PCC) after SARS-CoV-2 infection underscores the critical need for preparedness in addressing future post-acute infection syndromes (PAIS), particularly those linked to epidemic outbreaks. The lack of standardized clinical and epidemiological data during the COVID-19 pandemic has significantly hindered timely diagnosis and effective treatment of PCC, highlighting the necessity of pre-emptively standardizing data collection in clinical studies to better define and manage future PAIS. In response, the Cohort Coordination Board, a consortium of European-funded COVID-19 research projects, has reviewed data from PCC studies conducted by its members.

Background: Monitoring of somatic development through the assessment of anthropometric variables such as weight, height, and BMI is vital for evaluating the physical development and nutritional status of children. This approach aids in the early identification of somatic developmental disorders, enabling timely medical interventions. It traditionally relies on Z-scores, which compare anthropometric variables with reference standards.

Fine-flavored chocolates are distinguished by their complex and distinct flavor profiles, which includes notes such as floral, fruity, nutty, and spicy. This study sought to modulate the flavor development of chocolates by establishing controlled processing conditions during the transformation from seed to bean in a laboratory setting, to produce superior quality chocolates. Our experimental setup comprised two varying temperature levels (30 °C and 45 °C) and three organic acids (OAs: acetic, lactic, and citric acids) at concentrations of 1-30 g/L to adjust the pH of the transformation system.

Metabolic dysregulation, including perturbed glutamine-glutamate homeostasis, is common among patients with cardiovascular diseases, but the underlying mechanisms remain largely unknown. Using the human MESA cohort, here we show that plasma glutamine-glutamate ratio is an independent risk factor for carotid plaque progression. Mice deficient in glutaminase-2 (Gls2), the enzyme that mediates hepatic glutaminolysis, developed accelerated atherosclerosis and susceptibility to catastrophic cardiac events, while Gls2 overexpression partially protected from disease progression.

Article Synopsis
  • * A study analyzed LEPR variants in about 10,000 participants, identifying 86 rare heterozygous variants and confirming 12 as pathogenic, but most individuals with these variants did not show signs of obesity.
  • * The findings suggest no direct link between pathogenic LEPR variants and obesity risk or body mass index (BMI), raising concerns about prescribing setmelanotide based solely on the presence of these variants.

Summary: Phenotyping consists in applying algorithms to identify individuals associated with a specific, potentially complex, trait or condition, typically out of a collection of Electronic Health Records (EHRs). Because a lot of the clinical information of EHRs are lying in texts, phenotyping from text takes an important role in studies that rely on the secondary use of EHRs. However, the heterogeneity and highly specialized aspect of both the content and form of clinical texts makes this task particularly tedious, and is the source of time and cost constraints in observational studies.

Article Synopsis
  • Monocytes play a crucial role in atherosclerosis by turning into macrophages when they migrate to plaques, and this study explores how their glucose metabolism influences their behavior and contribution to the disease.
  • Researchers found that higher serum glucose levels are linked to increased monocyte numbers, while restricted diets hinder monocytes from switching energy sources, which reduces their presence in the blood.
  • The study highlights that glucose metabolism is vital for maintaining specific monocyte characteristics and functions, but inhibiting glucose uptake alone doesn't prevent atherosclerosis, likely because the remaining monocytes become more migratory.
