Background: There is growing evidence that epigenetic age acceleration may predict late life cognitive decline and dementia, but it is unknown whether this is due to accelerated neurodegeneration or reduction in cognitive resilience. We examined the relationship between epigenetic clocks and domain specific neuropsychological (NP) factor scores, mild cognitive impairment (MCI), Alzheimer's Disease (AD), and all-cause dementia, before and after accounting for plasma total tau (t-tau), a marker of neurodegeneration.

Method: DNA methylation and plasma t-tau (Simoa assay; Quanterix) data from 2091 Framingham Heart Study Offspring cohort participants were generated from blood at the same Exam 8 visit (2005-2008).

Clinical Manifestations.

Background: Subjective cognitive decline (SCD) is recognized to be in the Alzheimer's disease (AD) cognitive continuum. Aims: We examined the association between sex, age, temporal trends, and glycemic control among people with type 2 diabetes (T2D) in a multi-ethnic middle-income Asian country.

Methods: Using the National Diabetes Registry (2011-2020), we analyzed data for 221,769 adult Malaysians with T2D.We used quantile regressions to estimate the association of sex, age, and their interaction on HbAlevels at the 5th, 50th, and 95thpercentile and logistic regression to estimate the odds of good control (HbA < 7 %).

Reduced durability of hybrid immunity to SARS-CoV-2 in immunocompromised children.

Background: In endemic COVID-19, immunocompromised children are vulnerable until vaccinated but the optimal primary vaccination regime and need for booster doses remains uncertain.

Methods: We recruited 19 immunocompromised children (post-solid organ transplantation, have autoimmune disease or were on current or recent chemotherapy for acute lymphoblastic leukemia), and followed them from the start of primary vaccination with BNT162b2 mRNA SARS-CoV-2 until 1-year post-vaccination. We investigated the quality of vaccine immunogenicity, and longevity of hybrid immunity, in comparison to healthy children.

Management of operating theatre ambient temperature for major burn patients in a state-wide adult burns centre.

Introduction: The aim of this study was to investigate the efficacy of limiting increases in theatre ambient temperature to 27°C to prevent intraoperative patient hypothermia.

Methods: This single-centre, comparative cohort clinical study investigated the management of theatre ambient temperatures involving patients with ≥ 20 % TBSA burn injuries at Victorian Adult Burns Service (Melbourne, Australia). Data from the intervention group (August 2021 - February 2023, theatre ambient temperature increase limited to 27°C) was compared with a historical cohort (August 2019 - August 2021).

Hepatocytes are organized into distinct zonal subsets across the liver lobule, yet their contributions to liver homeostasis and regeneration remain controversial. Here, we developed multiple genetic lineage-tracing mouse models to systematically address this. Introduction: Family members caring for a person living with dementia (PWD) can experience caregiver burden, leading to psychological distress if unmanaged. It's essential for healthcare professionals, especially nurses to identify caregivers at risk of stress and depression, triggering prompt management during their contact with caregivers of PWD. The study team developed an evidence-based caregiver burden-mastery hybrid assessment and intervention decision matrix (CHAT-MI) for caregivers of PWD and examined its feasibility of use.

Estimated potassium intake and major adverse cardiovascular events in individuals with type 2 diabetes: a prospective cohort study with trans-ethnic validation.

Background: Data on the relationship between potassium intake and major cardiovascular events (MACE) in patients with diabetes are scarce. We aim to study the association between estimated potassium intake and risk of MACE in individuals with type 2 diabetes.

Here, we present a protocol to efficiently generate human artery and vein endothelial cells (ECs) from pluripotent stem cells within 3-4 days of differentiation. We delineate how to seed human pluripotent stem cells and sequentially differentiate them into primitive streak, lateral mesoderm, and either artery or vein ECs.

Cell- and tissue-specific glycosylation pathways informed by single-cell transcriptomics.

While single-cell studies have made significant impacts in various subfields of biology, they lag in the Glycosciences. To address this gap, we analyzed single-cell glycogene expressions in the Tabula Sapiens dataset of human tissues and cell types using a recent glycosylation-specific gene ontology (GlycoEnzOnto). At the median sequencing (count) depth, ∼40-50 out of 400 glycogenes were detected in individual cells.

Global prevalence of depression, anxiety, and stress symptoms in different trimesters of pregnancy: A meta-analysis and meta-regression.

The global prevalence of psychological problems in different trimesters is unclear due to methodological constraints in previous reviews. A precise estimate would be a key first step in raising awareness and allocating resources. This review aims to (1) calculate the global prevalence of depression, anxiety, and stress symptoms during different trimesters and (2) determine the factors influencing their prevalence estimates.
